    Uji Molisch merupakan uji umum untuk mendeteksi keberadaan karbohidrat dalam suatu sampel. Penelitian ini bertujuan untuk menerapkan uji Molisch pada beberapa sampel yang tidak diketahui kandungannya. Prinsip uji ini didasarkan pada reaksi dehidrasi karbohidrat oleh asam sulfat pekat menjadi furfural (untuk pentosa) atau hidroksimetilfurfural (untuk heksosa). Senyawa furfural kemudian bereaksi dengan α-naftol (pereaksi Molisch) membentuk senyawa kompleks berwarna ungu-keunguan. Hasil penelitian menunjukkan bahwa semua sampel karbohidrat (monosakarida, disakarida, dan polisakarida) memberikan hasil positif yang ditandai dengan terbentuknya cincin ungu pada antarmuka larutan. The reaction mechanism relies on the dehydration of carbohydrates to furfural (from pentoses) or hydroxymethylfurfural (from hexoses), which subsequently condenses with α-naphthol to form a colored complex. All carbohydrate samples (monosaccharides, disaccharides, and polysaccharides) yielded positive results, while non-carbohydrate controls remained negative. The test demonstrated high sensitivity and simplicity, confirming its utility as a preliminary screening method for carbohydrate analysis in biochemical research.
